Which of the following is desirable?

(a) Driver- driver hybrid

(b) Driver- tracer hybrid

(c) Tracer- tracer hybrid

(d) Driver alone

I have been asked this question by my school principal while I was bunking the class.

My question comes from Cloning the cDNA and Specialized Libraries topic in division Libraries Construction of Genetic Engineering

The correct option is (c) Tracer- tracer hybrid

Explanation: Our sequence of interest is present in the tracer. So if it hybridizes with a driver, it means that the sequence is present is it also and thus it is not required by us. Hence, either tracer-tracer hybrids or single stranded tracers are required.
